class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t<p>Sur cette th\u00e9matique de recherche nous d\u00e9veloppons des m\u00e9thodes de \u201cmonitoring\u201d ultrasonores pour le suivi de th\u00e9rapies induites par ultrasons. Ces combinaisons s\u2019appuient sur l\u2019utilisation de syst\u00e8mes multimodaux combinant les ultrasons \u00e0 vis\u00e9e th\u00e9rapeutique et une autre m\u00e9thode d\u2019imagerie.<\/p><h4>Les SUPERBHE<\/strong><\/h4>\nDepuis quelques ann\u00e9es, une technique d&rsquo;\u00e9chographie super-r\u00e9solue a \u00e9t\u00e9 propos\u00e9e afin de d\u00e9passer les limites de r\u00e9solution impos\u00e9es par l\u2019\u00e9chographie conventionnelle. Cette approche, nomm\u00e9e ULM, propose d\u2019exploiter les techniques de reconstruction utilis\u00e9es en imagerie optique. Ainsi, l\u2019image obtenue n\u2019est plus directement reconstruite \u00e0 partir du signal r\u00e9trodiffus\u00e9 par les microbulles mais divers algorithmes sont impl\u00e9ment\u00e9s afin de suivre le d\u00e9placement de ces agents pr\u00e9alablement inject\u00e9s dans le flux sanguin et reconstruire une cartographie du r\u00e9seau vasculaire avec une r\u00e9solution pouvant avoisiner les 5 \u00b5m. Selon la taille des vaisseaux et la vitesse du flux, le suivi du d\u00e9placement des microbulles peut n\u00e9cessiter l\u2019acquisition de plusieurs dizaines de milliers d\u2019images sur plusieurs secondes. Le d\u00e9veloppement de cette approche a \u00e9t\u00e9 possible gr\u00e2ce \u00e0 l\u2019imagerie en onde plane et \u00e0 l\u2019essor de nouveaux appareils \u00e9chographiques ultra-rapides au cours de ces derni\u00e8res ann\u00e9es.\u00a0Nous proposons ici d\u2019observer par \u00e9chographie super-r\u00e9solue les variations vasculaires induites par les ultrasons th\u00e9rapeutiques lors de technologies pour des applications th\u00e9ranostiques en imagerie combin\u00e9e PET \/ US en canc\u00e9rologie: Projet IMPETUS<\/h4><p>L&rsquo;imagerie mol\u00e9culaire fait partie de l\u2019arsenal th\u00e9rapeutiques des soins courants dans de nombreux cancers. En permettant aux m\u00e9decins de visualiser des processus internes au niveau cellulaire, elle fournit des informations aidant \u00e0 la d\u00e9tection, au traitement et au suivi du cancer. La tomographie par \u00e9mission de positrons (TEP) associ\u00e9e \u00e0 des radiotraceurs adapt\u00e9s est la technique de r\u00e9f\u00e9rence pour l\u2019imagerie fonctionnelle, m\u00e9tabolique et quantitative de biomarqueurs. L&rsquo;\u00e9chographie (US) est une modalit\u00e9 d\u2019imagerie temps-r\u00e9el fonctionnelle et anatomique pour le diagnostic. Sa capacit\u00e9 m\u00e9canique \u00e0 perm\u00e9abiliser les membranes joue \u00e9galement un r\u00f4le crucial en th\u00e9rapie par l&rsquo;am\u00e9lioration de la d\u00e9livrance de m\u00e9dicaments. IMPETUS vise \u00e0 \u00ab\u00a0synergiser\u00a0\u00bb ces deux modalit\u00e9s pour g\u00e9n\u00e9rer des innovations th\u00e9ranostiques en oncologie; des innovations en chimie de formulation des agents de contraste US (MB), ciblage et radiomarquage; des innovations technologiques avec de nouveaux dispositifs CMUT et des transducteurs TEP-compatibles pour monitorer et s\u00e9curiser les protocoles de perm\u00e9abilisation membranaire. Ensemble, ces d\u00e9veloppements feront de l&rsquo;imagerie mol\u00e9culaire quantitative US une r\u00e9alit\u00e9 gr\u00e2ce \u00e0 la combinaison de l&rsquo;imagerie TEP avec des \u00ab radioMB \u00bb. De plus, la d\u00e9livrance de m\u00e9dicaments par sono-perm\u00e9abilisation vers des tissus sanctuaris\u00e9s sera d\u00e9montr\u00e9e par la vectorisation de nanoparticules radiomarqu\u00e9es vers de tels tissus. Les preuves de concept seront \u00e9tablies sur un mod\u00e8le de glioblastome, l&rsquo;un des cancers les plus r\u00e9sistant \u00e0 toutes les th\u00e9rapies. Pour atteindre ces objectifs, un consortium de 3 laboratoires universitaires experts en radiochimie, physique, imagerie TEP et US (BioMaps), en formulation et d\u00e9livrance de MB (CBM) et en dispositifs et\u00a0technologies US (GREMAN) a \u00e9t\u00e9 constitu\u00e9. Les ultrasons (US) associ\u00e9s \u00e0 l\u2019injection de microbulles gazeuses apportent une solution prometteuse en perm\u00e9abilisant localement et transitoirement cette barri\u00e8re. Dans le cadre de la translation clinique de cette technologie, la dosim\u00e9trie du champ acoustique \u00e0 travers le cr\u00e2ne humain doit \u00eatre ma\u00eetris\u00e9e en temps r\u00e9el pour assurer l\u2019efficacit\u00e9 et la s\u00e9curit\u00e9 du protocole. L\u2019intensit\u00e9 US doit \u00eatre suffisamment importante pour induire une vibration des microbulles agissant sur la perm\u00e9abilit\u00e9 des vaisseaux sanguins tout en restant suffisamment mod\u00e9r\u00e9e afin d\u2019\u00e9viter une implosion de ces microbulles induisant des dommages permanents. Ce projet vise \u00e0 am\u00e9liorer l\u2019exploitation du signal de cavitation en proposant l\u2019\u00e9valuation de capteurs US innovants, l\u2019optimisation de leur positionnement sur le cr\u00e2ne du patient et l\u2019impl\u00e9mentation d\u2019une technique d\u2019analyse ultra-rapide de la r\u00e9ponse des microbulles. href=\"https:\/\/phase1.attract-eu.com\/showroom\/project\/echobrain\/\" target=\"_blank\" rel=\"noopener\">ECHOBRAIN<\/a><\/h4><p>Le projet de recherche Echobrain vise \u00e0 innover en imagerie par ultrasons transcr\u00e2niens (US). En effet, l&rsquo;\u00e9chographie c\u00e9r\u00e9brale pourrait \u00eatre beaucoup plus accessible que les techniques actuellement utilis\u00e9es, telles que les rayons X ou l&rsquo;imagerie par r\u00e9sonance magn\u00e9tique (IRM), mais reste difficile en raison de la forte att\u00e9nuation et de l&rsquo;aberration du faisceau US lors de la travers\u00e9e de l&rsquo;os du cr\u00e2ne. La qualit\u00e9 de l&rsquo;\u00e9chographie transcr\u00e2nienne peut \u00eatre consid\u00e9rablement augment\u00e9e \u00e0 condition que les distorsions du cr\u00e2ne puissent \u00eatre prises en compte \u00e0 l&rsquo;\u00e9mission, pour focaliser le faisceau \u00e0 l&rsquo;int\u00e9rieur du cerveau; et \u00e0 la r\u00e9ception, pendant le processus de formation de l&rsquo;image. Ces distorsions peuvent \u00eatre mesur\u00e9es exp\u00e9rimentalement, mais n\u00e9cessitent une proc\u00e9dure invasive utilisant g\u00e9n\u00e9ralement des sondes situ\u00e9es \u00e0 l&rsquo;int\u00e9rieur de la zone \u00e0 imager. Des mod\u00e8les de propagation des ondes US, utilisant une description de la morphologie du cr\u00e2ne obtenue par IRM ou rayons X peuvent \u00e9galement \u00eatre utilis\u00e9s pour pr\u00e9dire ces corrections d&rsquo;aberrations de mani\u00e8re non invasive. Ce projet se focalise sur une m\u00e9thode de propagation simplifi\u00e9e suffisante pour obtenir une correction satisfaisante du cr\u00e2ne. D&rsquo;un point de vue technologique, il contribuera au d\u00e9veloppement de nouveaux scanners US d\u00e9di\u00e9s \u00e0 l&rsquo;imagerie c\u00e9r\u00e9brale bas\u00e9s sur une \u00e9lectronique ultrarapide programmable. D&rsquo;un point de vue m\u00e9dical, pratiquement toutes les maladies du cerveau pourraient b\u00e9n\u00e9ficier de cette am\u00e9lioration.
