restent peu efficaces pour le traitement de certaines pathologies. Cette m\u00e9thode non invasive repose sur les oscillations des microbulles induites par les ultrasons \u00e0 proximit\u00e9 de barri\u00e8res biologiques afin d&rsquo;augmenter transitoirement leur perm\u00e9abilit\u00e9. Cette strat\u00e9gie peut \u00eatre notamment utilis\u00e9e \u00e0 travers le cr\u00e2ne pour la d\u00e9livrance cibl\u00e9e de mol\u00e9cules incluant des chimioth\u00e9rapies ou des anticorps monoclonaux. Cependant, l\u2019efficacit\u00e9 du traitement ultrasonore reste limit\u00e9e par la faible dur\u00e9e de vie (quelques minutes) et la taille des microbulles (agents intravasculaires). L\u2019utilisation de nanogouttelettes pourrait augmenter significativement la d\u00e9livrance de m\u00e9dicaments dans le cerveau. Nous travaillons sur la formulation et la caract\u00e9risation de ces nouveaux agents, \u00e0 la fois stables dans le sang et sensibles aux ultrasons, pour la th\u00e9rapie du cerveau.<\/p>\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<section class=\"elementor-section elementor-inner-section elementor-element elementor-element-9aa6c44 elementor-section-boxed sono-perm\u00e9abilisation n\u00e9cessite une ma\u00eetrise en temps r\u00e9el de la dosim\u00e9trie ultrasonore pour assurer l\u2019efficacit\u00e9 et la s\u00e9curit\u00e9 du protocole. En effet, l\u2019oscillation non contr\u00f4l\u00e9e des bulles peut induire des dommages irr\u00e9versibles aux tissus. Ce contr\u00f4le peut \u00eatre assur\u00e9 \u00e0 l\u2019aide de transducteurs ultrasonores passifs dispos\u00e9s autour de la zone \u00e0 traiter. Dans ce projet, nous proposons de d\u00e9velopper de nouveaux capteurs (cMUT) afin de d\u00e9tecter le signal de cavitation lors de protocoles de th\u00e9rapie par ultrasons. Les cMUT offrent de nombreux avantages tels qu\u2019une r\u00e9ponse fr\u00e9quentielle plus large et une plus grande facilit\u00e9 d\u2019int\u00e9gration \u00e9lectronique. Avec ces capteurs, nous souhaitons donc am\u00e9liorer la d\u00e9tection des bulles et d\u00e9finir en temps r\u00e9el les doses de cavitation n\u00e9cessaires \u00e0 une perm\u00e9abilisation du cerveau : projet DELIVUS<\/h4><p>Depuis quelques ann\u00e9es, il existe un int\u00e9r\u00eat grandissant sur l\u2019utilisation des immunoth\u00e9rapies pour lutter contre le cancer. Bien que ces th\u00e9rapies aient montr\u00e9 des r\u00e9sultats prometteurs dans le traitement de plusieurs cancers p\u00e9riph\u00e9riques, l\u2019efficacit\u00e9 reste limit\u00e9e pour les tumeurs du cerveau. En effet, la pr\u00e9sence de la barri\u00e8re h\u00e9mato-enc\u00e9phalique (BHE), prot\u00e9geant le cerveau, limite drastiquement le passage des anticorps du sang vers la tumeur c\u00e9r\u00e9brale. Nous proposons donc d\u2019utiliser la technique de sono-perm\u00e9abilisation pour favoriser leur d\u00e9livrance dans le cerveau et traiter le cancer (en partenariat avec C. Truillet, IC CEA, eq. 3 BioMaps). Nous travaillons actuellement sur le d\u00e9veloppement de s\u00e9quences ultrasonores appliqu\u00e9es \u00e0 travers le cr\u00e2ne permettant une d\u00e9livrance homog\u00e8ne, reproductible et combin\u00e9e aux ultrasons : projet RIVUS<\/h4><p>La radioth\u00e9rapie interne vectoris\u00e9e (RIV) consiste \u00e0 injecter au patient un radioth\u00e9rapeutique distribu\u00e9 dans tout l\u2019organisme avec une accumulation pr\u00e9f\u00e9rentielle au niveau des cellules tumorales. Cette m\u00e9thode permet de d\u00e9livrer localement un rayonnement de haute \u00e9nergie induisant des d\u00e9g\u00e2ts irr\u00e9versibles et menant \u00e0 la destruction de la tumeur. Malgr\u00e9 des r\u00e9sultats tr\u00e8s prometteurs, l\u2019utilisation de la RIV est r\u00e9serv\u00e9e \u00e0 des patients \u00e0 un stade avanc\u00e9. En effet, l\u2019accumulation non n\u00e9gligeable d\u2019agents radioactifs dans les reins ou le foie induit une toxicit\u00e9 importante impactant le bien-\u00eatre du patient. Nous proposons ici d\u2019utiliser la sono-perm\u00e9abilisation afin de favoriser le passage de mol\u00e9cules du sang vers la tumeur et d\u2019induire une accumulation plus sp\u00e9cifique de l\u2019agent radioactif dans la tumeur.
